OBJECTIVE: This study compared the protective effect of an experimental TiF4/Chitosan toothpaste with a commercial toothpaste on the prevention of erosive tooth wear (ETW) in situ. METHODS: Fifteen subjects took part in this crossover and double-blind study, in which they wore a palatal appliance containing 4 bovine enamel and 4 dentin in 3 phases (5 days each). Half of the samples were subjected to erosive challenges (90 s in 0.1 % citric acid, pH 2.5, 4 times/day), and the other half to erosive plus abrasive challenges (15 s plus 45 s of contact, 2 times/day). The phases corresponded to the application of the different toothpastes: 1) TiF4 (1400 ppm F-) plus Chitosan, 2) Elmex®, Erosion Protection (1400 ppm F-, Chitosan), and 3) Placebo (negative control). Tooth wear was measured using contact profilometry (µm) and submitted to two-way RM ANOVA/Tukey test (p < 0.05). RESULTS: No significant differences were detected between the experimental and commercial toothpastes, regardless of the challenge on both tissues. Both significantly reduce ETW compared to negative control (p < 0.0006). Tooth wear was increased by brushing only on eroded enamel (p < 0.01), but not on dentin (p = 0.6085). TiF4/Chitosan [erosion 2.98 ± 1.12 µm vs. erosion and abrasion 3.12 ± 1.33 µm] and Elmex® toothpastes [erosion 2.35 ± 0.93 µm vs. erosion and abrasion 2.98 ± 1.0 µm] minimized the impact of brushing compared to placebo on enamel [erosion 4.62 ± 1.48 µm vs. erosion and abrasion 5.15 ± 1.50 µm]. CONCLUSIONS: TiF4 plus chitosan toothpastes showed to be effective in minimizing the ETW as the commercial toothpaste is in situ. CLINICAL RELEVANCE: The experimental toothpaste has similar effect against ETW compared to the commercial toothpaste. Considering the increased ETW prevalence worldwide, this result supports clinical trials and a possible application of this experimental anti-erosive toothpaste in the future.

The effect of solutions containing a statherin-derived peptide (Stn15pSpS) on the protection against enamel erosion in vitro was evaluated. Bovine enamel specimens were divided into 4 groups (n = 15/group): (1) deionized water (negative control), (2) Elmex Erosion Protection™ (positive control), (3) 1.88 × 10-5 M Stn15pSpS, and (4) 3.76 × 10-5 M Stn15pSpS. The solutions were applied on the specimens for 1 min. Stimulated saliva was collected from 3 donors and used to form a 2-h acquired pellicle on the specimens. Then, the specimens were submitted to an erosive pH-cycling protocol 4 times/day, for 7 days (0.01 M HCl pH 2.0/45 s, artificial saliva/2 h, and artificial saliva overnight). The solutions were applied again during pH-cycling, 2 times/day for 1 min after the first and last erosive challenges. Enamel loss (µm) was assessed by contact profilometry. Data were analyzed by Kruskal-Wallis and Dunn's test (p < 0.05). The best protection against erosion was conferred by Elmex Erosion Protection that significantly differed from all the other treatments, followed by the solutions containing Stn15pSpS, regardless of the concentration. However, 3.76 × 10-5 M Stn15pSpS did not differ from the negative control. The solution containing the lower concentration of Stn15pSpS protected against erosion in vitro, which should be confirmed using protocols that more closely resemble the clinical condition.

The effect of gels containing a statherin-derived peptide (Stn) on the protection against enamel and dentin erosive tooth wear (ETW) in vitro was evaluated. Bovine enamel and dentin specimens were divided into 2 groups (n = 15 and 18/group for enamel and dentin, respectively) that were treated with Chitosan or Carboxymethylcellulose (CMC) gels containing Stn15pSpS at 1.88 × 10-5 M or 3.76 × 10-5 M. Chitosan or CMC gels without active ingredients served as negative controls, while chitosan gel containing 1.23% F (as NaF) and acidulated phosphate fluoride gel (1.23% F) served as positive controls. The gels were applied on the specimens for 4 min. Stimulated saliva was collected from 3 donors and used to form a 2-h acquired pellicle on the specimens. Then, the specimens were submitted to an erosive pH cycling protocol 4 times/day for 7 days (0.01 M HCl pH 2.0/45 s, artificial saliva/2 h, and artificial saliva overnight). The gels were applied again during pH cycling, 2 times/day for 4 min after the first and last erosive challenges. Enamel and dentin loss (µm) were assessed by contact profilometry. Scanning electron microscopy (SEM) was analyzed using a cold field emission. Data were analyzed by two-way ANOVA (for chitosan and CMC gels, separately) and Tukey's multiple comparison test. SEM images showed changes to enamel topography after application oft the gels containing Stn or F. Regarding CMC-based gels, for enamel, none of the treatments significantly reduced ETW in comparison with placebo; for dentin, however, gels containing Stn, regardless the concentration, significantly reduced the ETW. Moreover, Chitosan-based gels, regardless the Stn concentration, were able to protect enamel and dentin against ETW. Gels containing Stn might be a new approach to protect against ETW.

This study evaluated the protective effect of TiF4 and chitosan toothpaste on erosive tooth wear (ETW) in vitro. Enamel and dentin samples were randomly assigned to toothpastes (n = 12): (G1) TiF4 (1400 ppm F-), (G2) 0.5% chitosan (75% deacetylation, 500 mPas), (G3) TiF4 (1400 ppm F-) plus 0.5% chitosan (75% deacetylation, 500 mPas), (G4) Placebo, (G5) Erosion Protection (Elmex-GABA, 1400 ppm F-). Twelve samples were only eroded. All samples were submitted to erosive pH cycles and G1 to G5 to abrasive challenges using toothpastes' slurries plus 45 s of treatment, for 7 days. The final profile was overlaid to the baseline one for the ETW calculation (µm). The data were subjected to Kruskal-Wallis/Dunn tests. TiF4 toothpastes, regardless of the presence of chitosan, were able to significantly reduce ETW compared to placebo, while chitosan alone was similar to placebo for both tissues. The toothpastes containing TiF4 were even superior to the commercial Elmex toothpaste on enamel, while they were similar on dentin; both were also significantly different from placebo for both tissues. TiF4 and Elmex toothpastes minimized the impact of brushing on eroded surface. In conclusion, TiF4 toothpastes, regardless the presence of chitosan, showed to be effective in minimizing ETW in vitro.

Avaliou-se a interferência das nanopartículas de prata sobre a angiogênese relacionada ao crescimento tumoral. A pesquisa científica foi realizada através da incubação de 42 ovos embrionados de galinhas. Após 24 horas de incubação, esses mesmos ovos foram separados em seis grupos contendo sete ovos cada, para os tratamentos com: Grupo 1: Soro fisiológico; Grupo 2: Tumor de Ehrlich (TE); Grupo 3: Nanopartículas de prata; Grupo 4: Prednisolona; Grupo 5: Nanopartículas de prata e Tumor de Ehrlich; Grupo 6: Prednisolona e Tumor de Ehrlich. Após o tempo total de incubação, as membranas corioalantoideas (MCAs) foram removidas, e analisadas através do microscópio de luz e fotografadas. O grupo 1 apresentou um padrão normal de crescimento e foi utilizado como controle negativo; O grupo 2 apresentou um aumento na quantidade de vasos sanguíneos; o grupo 3 apresentou baixa interferência na angiogenese embrionária e não contribuiu para o desenvolvimento do tumor; O grupo 4 demonstrou diminuição no desenvolvimento de vasos sanguíneos; O grupo 5 indicou que as nanopartículas de prata, quando associadas ao TE, não favorece o desenvolvimento tumoral e o grupo 6 demonstrou que o fármaco prednisolona associado ao TE, se comporta como um excelente inibidor de neoangiogese tumoral. Considera-se através da técnica realizada a possibilidade de utilizar nanopartículas de prata para o tratamento de células tumorais de Ehrlich, porém devem ser realizados testes confirmatórios para estudar a relação da substância descrita às células tumorais empregadas.


The interference of silver nanoparticles on angiogenesis related to tumor growth was evaluated. Scientific research was carried out by incubating 42 embryonated chicken eggs. After 24 hours of incubation, these same eggs were separated into six groups containing seven eggs each, for treatments with: Group 1: Saline; Group 2: Ehrlich's tumor (ET); Group 3: Silver nanoparticles; Group 4: Prednisolone; Group 5: Silver nanoparticles and Ehrlich's Tumor; Group 6: Prednisolone and Ehrlich's Tumor. After the total incubation time, the chorioallantoid membranes (MCAs) were removed, and analyzed using a light microscope and photographed. Group 1 showed a normal growth pattern and was used as a negative control; Group 2 showed an increase in the amount of blood vessels; group 3 showed low interference in embryonic angiogenesis and did not contribute to the development of the tumor; Group 4 demonstrated a decrease in the development of blood vessels; Group 5 indicated that silver nanoparticles, when associated with TE, do not favor tumor development and group 6 demonstrated that the drug prednisolone associated with TE, behaves as an excellent inhibitor of tumor neoangiogenesis. Through the technique performed, the possibility of using silver nanoparticles for the treatment of Ehrlich tumor cells is considered, however, confirmatory tests should be performed to study the relationship of the substance described to the tumor cells employed.

Introdução: O vírus SARS-CoV-2, causador da COVID-19, tornou- -se a representação de ameaça global ao que diz respeito à saúde pública, à medida que se espalha facilmente. A COVID-19 pode gerar desde um quadro assintomático até sinais e sintomas envolvendo os sistemas respiratório, hepático, neurológico, podendo, inclusive, levar à morte. O sistema imunológico possui papel fundamental no combate às infecções e sua atuação pode definir o quadro clínico, seja assintomático, leve, grave ou fatal. Objetivo: Descrever o papel das respostas imunológicas inata e adaptativa ao SARS-CoV-2. Metodologia: O estudo caracterizou-se por levantamento bibliográfico utilizando-se artigos científicos indexados nas bases de dados online PubMED, Medline, Scielo e Google Acadêmico, no período compreendido entre 2019 e 2020 Para a filtragem das publicações foram utilizados os seguintes descritores em português/inglês: COVID-19, SARS-CoV-2, Sistema imunológico, Resposta inata e Resposta adaptativa. Após filtragem, 42 artigos científicos foram selecionados. Resultados: O SARS-CoV-2 infecta as células do indivíduo ao reconhecer a enzima ECA2 ou a proteína CD147 e suas proteínas virais prejudicam a resposta imune à medida que diminuem a síntese de IFN1, ativam a via NF-kB, aumentam a síntese de citocinas pró-inflamatórias e aumentam a necroptose. Linfócitos T CD4+ possuem papel essencial na ativação de Linfócitos B e sua consequente síntese de anticorpos monoclonais. Linfócitos T CD8+ citotóxicos são capazes de dizimar células infectadas. Entretanto, estes eventos podem levar à resposta inflamatória exacerbada e à tempestade de citocinas, o que pode ser prejudicial nas infecções por SARS- -CoV-2, já que agravam sintomas e promovem alterações sistêmicas. Conclusão: Conhecido que Linfócitos T e B são ativados por peptídeos virais e que esta ativação promove a liberação de mediadores inflamatórios-chave e anticorpos monoclonais, e sabendo-se que a tempestade de citocinas inflamatórias pode agravar o quadro clínico do paciente infectado, faz-se necessário reconhecer a importância do sistema imunológico no combate à infecção da COVID-19.


Introduction: The SARS-CoV-2 virus, which causes COVID-19, has become the representation of global threat to public health, as it spreads easily, without efficient resources to control its spread until the present moment. COVID-19 can generate from an asymptomatic condition to signs and symptoms involving the respiratory, hepatic, neurological systems, and can even lead to death. The immune system has a fundamental role in fighting infections and its performance can define the clinical condition, either asymptomatic, mild, severe or fatal. Objective: To describe the role of innate and adaptive immune responses to SARS-CoV-2. Methodology: The study was characterized by a bibliographic survey, using scientific articles indexed in the online databases PubMED, Medline, Scielo and Google Scholar, in the period between 2019 and 2020. For filtering publications, the following descriptors were used in Portuguese/ English COVID-19, SARS-CoV-2, Immune system, Innate response and Adaptative response. After filtering, 42 scientific articles were selected. Results: SARS-CoV-2 infects the individual's cells by recognizing the ECA2 enzyme or the CD147 protein and its viral proteins impair the immune response as they decrease IFN1 synthesis, activate the NF-kB pathway, increase pro cytokine synthesis -inflammatory and increase necroptosis. CD4 + T lymphocytes play an essential role in the activation of B lymphocytes and their consequent synthesis of monoclonal antibodies. Cytotoxic CD8 + T lymphocytes are capable of decimating infected cells. However, these events can lead to an exacerbated inflammatory response and a cytokine storm, which can be harmful in SARS-CoV-2 infections, as they worsen symptoms and promote systemic changes. Conclusion: Known that T and B lymphocytes are activated by viral peptides and that this activation promotes the release of key inflammatory mediators and monoclonal antibodies, and knowing that the storm of inflammatory cytokines can worsen the clinical condition of the infected patient, it is necessary to recognize the importance of the immune system in combating the infection of COVID-19.
